What is the air vent/nozzle is for?

Although it resembles the drinking tip on some sports drink and water bottles, it should not be used for that purpose. It is designed to allow for the displacement of air from the empty bottle as water enters through the filter element. If the air vent remains closed while filling the bottle, water would still pass through the element but would be impeded and slowed down significantly as the air inside of the bottle would be compressed and resist the entering water. With the pour through method of filling a bottle you will not observe the air coming out, but with the immersing method, there will be bubbles. The escaping air prevents any water from entering through this opening. ~ David

A 40 X 40 MICRON mesh can filter out what size micron particle?

A standard microfilter will filter out particulate greater than or equal to 100 nm. Based on other products this should get rid of 2.4-3LRV of bacteria (99%-99.9%). Your main aim is getting rid of turbidity (floaties) otherwise the units UV won't penetrate everywhere. They're a great little supplement but they're not made for any nasties or you'd need more than gravity to get the water through
